Hey!
is there any way to make arrows that have/are:
– Different color of core versus edge (for example red edge with yellow fill/core?
– Different colour of arrow-head than of the actual arrow-line?
– Gradient from beginning to end, or reverse?

Just to clarify, I assume you're mean for lines drawn with the line tool with arrowheads/tails, right? (Versus one of the arrow shapes from a palette that has a fill of some sort applied?)
OmniGraffle doesn't currently support that, but we do have a feature request open. We generally try to assign popular requests higher priority, so if you email the support ninjas (omnigraffle@omnigroup.com) and let them know you need this, they can add you to the request database.
In the short term, if you can live with straight lines, you might be able to cobble something together by drawing really narrow rectangles for the body of the arrow, then unioning that with a triangle for the head...
