Is it possible to create a shape with a line connector as indicated in the example image below so, as in this example, the callout box can attach itself to magnets on other shapes?

Magnets only interact with lines, in their current implementation.
You could make a suggestion through Help → Send Feedback in the main OmniGraffle menu, but in the meanwhile, grouping is the only option that comes to mind.
