Yes. In the Style inspector (command-1), click on the second tab (Lines and Shapes) while you have a line selected and you'll get options for line endings, thickness, corner radii, etc.
Definitely take the time to watch the videos on the OmniGraffle product page! After watching them, try making some diagrams, then watch the videos again. There's a lot of information to be absorbed.
